Plasmid for the expression of a Spinach2 aptamer flanked by a tRNALys3 scaffold under the control of T7 RNA polymerase. The plasmid is suitable for transcription both in vitro and in vivo in E. coli strains carrying the T7 RNA polymerase gene. Spinach2 is an improved version of the fluorogenic RNA-aptamer spinach developed by Strack et al. for live imaging of cells (doi.org/10.1038/nmeth.2701). The aptamer provides fluorescence in presence of the fluorophore DFHBI or derivatives (excitation/emission wavelength: 447/501 nm).
