Plasmid for the expression of an iSpinach aptamer flanked by a tRNALys3 scaffold under the control of T7 RNA polymerase. The plasmid is suitable for transcription both in vitro and in vivo in E. coli strains carrying the T7 RNA polymerase gene. iSpinach is an improved version of the fluorogenic RNA-aptamer spinach far less salt-sensitive and with a much higher thermal stability and it was developed by Autor et al. (doi.org/10.1093/nar/gkw083). The aptamer provides fluorescence in presence of the fluorophore DFHBI or derivatives (excitation/emission wavelength: 447/501 nm).
