Plasmid for the expression of a orange Broccoli aptamer flanked by a tRNALys3 scaffold under the control of T7 RNA polymerase. The plasmid is suitable for transcription both in vitro and in vivo in E. coli strains carrying the T7 RNA polymerase gene. oBroccoli is a fluorogenic RNA aptamer developed by Song et al. for live imaging of cells (doi.org/10.1038/nchembio.2477). The aptamer provides fluorescence in presence of the fluorophore DFHO (excitation/emission wavelength: 505/545 nm).
