CIDAR MoClo relies upon Type IIS restriction enzymes that leave specific 4bp overhangs, called hinges, that are used for parts assembly. CIDAR MoClo relies on 8 hinges specific to each part. For example, coding sequence (CDS) parts are flanked with AATG (abbreviated C) at the 5′ end and ATTG (abbreviated D) on the 3′ end, making all CDS parts
interchangeable.
