DLX_PmeR-Ppme_v3.0

The biomodule carries all the elements to drive RNA transcription under the control of phloretin in E. coli. An operator for the TetR-family factor PmeR was evolved to allow the induction of gene expression by phloretin (doi.org/10.1093/nar/gkz772). The coding sequence of the PmeR repressor is driven by a constitutive promoter directed in the opposite direction of the phloretin-inducible promoter Ppme. PmeR gene transcription is interrupted by a transcription terminator and PmeR coding sequence was optimized to remove all the restriction sites incompatible with DNAmate assembly. The biomodule shall be placed right upstream of a 5' UTR sequence of a gene of interest to have the expression controlled by phloretin.
