MIAMI - Standard template for Terminator testing
This construct is the standard template to test Terminator's strength in vivo in S.cerevisiae [1] developed by the MIAMI consortium using the protocol described in [2]. Briefly, the parts can be cloned into the appropriate placeholder of the MIAMI – Standard reporter gene cassette [3] and integrated into S.cerevisiae genome by EasyClone marker-free integration as described in [4]. Terminator's strength is then assessed in vivo over 24 hours and results are re-scaled using the standard operating procedure described in [5].
